11β-HSD1参与海马和下丘脑-垂体-肾上腺轴的神经内分泌调节

摘要:

11β-羟类固醇脱氢酶1型(11β-hydroxysteroid de-hydrogenase type 1,11β-HSD1)是糖皮质激素(glu-cocorticoid,GC)代谢酶,调节局部组织中有活性的糖皮质激素水平。下丘脑-垂体-肾上腺轴(hypothalam ic-pituitarya-drenal axis,HPA)是调控GC的重要反馈轴;而海马是调节HPA功能的重要中枢组织,也是与认知、学习、记忆功能密切相关的脑区,海马受高组织水平GC的影响可引起年龄相关的认知功能障碍。中枢部位11β-HSD1对海马、HPA的影响引起诸多学者的关注,作者就这方面的研究进展作一综述。1 11β-HSD1的理化特性及生物学特性1.1概述11β-HSD1是一种NADP(H)依赖性酶,属断链脱氢/还原酶蛋白超家族成员,肝、肺、脂肪组织、骨、生殖腺和大脑均有不同程度的表达[1]。11β-HSD1表达水平受多种因素相互影响与调节。皮下及网膜脂肪细胞经肿瘤坏死因子-α(tumor necrosis factor-,αTNF-α)、白介素-1b、瘦素等作用后,11β-HSD1表达呈剂量依赖性增加;胰岛素样生长因子-1作用后,...
