结缔组织生长因子单克隆抗体超微超顺磁氧化铁粒子的制备及其生物活性的研究

摘要:

目的:通过化学合成方法制作结缔组织生长因子(connective tissue growth factor,CTGF)单克隆抗体超微超顺磁氧化铁(ultrasmall superparamagnetic iron oxide,USPIO)粒子分子探针,并检测其生物活性。方法:采用EDC(1-ethyl-3-[3-dimethylamin-opropyl]carbodiimide hydrochloride)化学连接剂将CTGF单克隆抗体与DMSA[meso-2,3-dimercaptosuccinic acid(HOOCCH(SH)-CH(SH)-COOH)]修饰的USPIO相结合,形成具有免疫活性的分子探针。通过间接酶联免疫吸附试验(ELISA)及Western blot-ting方法检测其生物稳定性及免疫活性。结果:随着加入不同质量的单克隆抗体,ELISA试验所得的吸光度值随抗体含量增加而增大,与阴性对照USPIO组相比较差异显著(P<0.05),分别间隔30 d及60 d后再次测量,只有200μg单抗组吸光度值降低(P<0.05)。Western blotting试验显示,结合单抗的USPIO及单纯单抗...
