骨髓间充质干细胞对小鼠肉瘤细胞S180生长的影响

摘要:

目的:研究骨髓间充质干细胞(BMSCs)对小鼠肉瘤细胞S180生长的影响,并探讨其可能的机制。方法:采用密度梯度离心法结合贴壁培养法分离、纯化、扩增小鼠BMSCs,流式细胞术检测其细胞表面分子的表达。收集小鼠BMSCs上清液,采用MTT法检测其对小鼠S180细胞的生长抑制效应;流式细胞术检测细胞凋亡率;RT-PCR检测p53、bc l-2 mRNA的表达情况。结果:密度梯度离心法结合贴壁培养法能有效分离、纯化小鼠BMSCs,流式细胞术分析结果显示培养的小鼠BMSCs阳性表达CD44(86.53%)、CD29(95.27%),阴性表达CD34(3.57%)。MTT法结果显示,小鼠BMSCs上清液可明显抑制S180细胞的生长,呈时间-剂量依赖性;经40%、80%上清液作用48 h后的凋亡率分别为14.17%、29.77%,明显高于对照组的4.23%(均P<0.01);p53及bc l2-mRNA均表达,小鼠BMSCs上清液可上调p53 mRNA表达,下调bc l2-mRNA表达。结论:小鼠BMSCs上清液可抑制小鼠S180细胞的生长并诱导细胞凋亡,其机制可能与上调p53、下调bc l-2基因表达有关。
