垂盆草醇提物对人肝癌细胞HepG2的抑制作用及其机制初探

摘要:

目的:观察垂盆草醇提物(CCW)对人肝癌细胞HepG2生长及其肿瘤相关分子指标的影响。方法:MTT法测定CCW对HepG2细胞增殖的作用;流式细胞术观察细胞周期的改变;分别用RT-PCR及免疫细胞化学法检测CCW对VEGF、c-Myc基因及蛋白水平的影响。结果:HepG2细胞的增殖在CCW作用的第24、48和72 h均被显著地抑制;CCW(100、200μg.m l-1)可使HepG2细胞阻滞于G0/G1期;CCW可显著下调VEGF、c-Myc的mRNA及蛋白水平的表达。结论:CCW对HepG2细胞增殖具有明显的抑制作用,并且能阻止细胞进入G2/M期,这可能与c-Myc基因表达的下调密切相关;CCW能抑制HepG2细胞的VEGF分泌,初步推测CCW具有抗血管生成作用。
