胃癌组织中HEG1、KRIT1的表达及其临床意义

摘要:

目的: 分析胃癌患者癌组织中具有表皮生长因子(EGF)样结构域的心脏发育蛋白1(HEG1)、Krev相互作用捕获蛋白1(KRIT1)的表达及其临床意义。方法: 选取我院2014年9月至2016年9月收治的胃癌患者85例,术中收集胃癌组织85例以及癌旁正常黏膜组织57例。采用免疫组化法检测组织中HEG1、KRIT1蛋白表达,分析HEG1、KRIT1蛋白水平与患者临床病理特征及预后的关系,并通过Ualcan数据库分析HEG1、KRIT1 mRNA在胃癌组织中的表达及其与胃癌患者总生存率的关系;Cox回归分析影响胃癌患者预后的因素。结果: Ualcan数据库显示,胃癌组织中HEG1、KRIT1 mRNA水平均高于正常黏膜组织(P<0.05),HEG1 mRNA高表达患者总生存率低于低表达患者(P<0.05);胃癌组织中HEG1、KRIT1蛋白阳性率均高于癌旁正常黏膜组织(P<0.05),胃癌组织中HEG1、KRIT1蛋白阳性率与患者胃癌分化程度、浸润深度、淋巴结转移有关(P<0.05),HEG1、KRIT1蛋白阳性患者5年生存率均低于阴性患者(P<0.05),且二者同时表达阳性的患者生存率更低(P<0.05)。肿瘤低分化、有淋巴结转移、HEG1及KRIT1蛋白阳性是影响胃癌患者预后的独立危险因素(P<0.05)。结论: 胃癌组织中HEG1、KRIT1均呈高表达,二者水平可预测患者预后。

Objective: To analyze the expression and clinical significance of heart development protein with epidermal growth factor(EGF)-like domains 1(HEG1) and Krev interaction trapped protein 1(KRIT1) in cancer tissues of gastric cancer patients. Methods: A total of 85 patients with gastric cancer admitted to our hospital from September 2014 to September 2016 were selected, and 85 cases of gastric cancer tissues and 57 cases of normal mucosal tissues adjacent to the cancer were collected intraoperatively. Immunohistochemical method was used to detect the expression of HEG1 and KRIT1 proteins in the tissues, and the relationship between the HEG1 and KRIT1 protein levels and the clinicopathological characteristics and prognosis of gastric cancer patients was analyzed. The Ualcan database was used to analyze the expression of HEG1 and KRIT1 mRNAs in gastric cancer tissue and their relationship with the overall survival rate of gastric cancer patients. Cox regression analysis was conducted to identify the factors affecting the prognosis of the patients. Results: Data from the Ualcan database showed that the levels of HEG1 and KRIT1 mRNAs in gastric cancer tissues were higher than those in normal mucosal tissues(P<0.05), and the overall survival rate of patients with high HEG1 mRNA expression was lower than that of patients with low expression(P<0.05). The positive rates of HEG1 and KRIT1 proteins in gastric cancer tissues were higher than those in normal mucosal tissues adjacent to cancer(P<0.05), and were associated with the degree of differentiation, depth of invasion and lymph node metastasis of gastric cancer(P<0.05). The 5-year survival rates of HEG1 and KRIT1 protein-positive patients were lower than those of HEG1 and KRIT1 protein-negative patients(P<0.05), and the patients with both positive expressions had an even lower survival rate(P<0.05). Low degree of cancer differentiation, lymph node metastasis, HEG1 and KRIT1 protein positive were independent risk factors affecting the prognosis of gastric cancer patients(P<0.05). Conclusion: Both HEG1 and KRIT1 levels are highly expressed in gastric cancer tissues, which can be used to predict the prognosis of patients.
