miR-105联合MMP-2预测脑胶质瘤术后复发的临床价值分析

摘要:

目的:探讨基质金属蛋白酶-2(MMP-2)与微小RNA (miR)-105对胶质瘤细胞侵袭、迁移和凋亡的影响,及二者预测术后复发的价值。方法:选取2018年7月至2021年7月我院手术切除的89例脑胶质瘤组织及配对的瘤旁组织,RT-PCR检测miR-105表达水平;免疫组化法检测MMP-2表达。用Logistic和受试者工作特征(ROC)曲线分析MMP-2、miR-105预测脑胶质瘤术后复发的价值。选取脑胶质瘤细胞株U251,分别转染空白质粒、miR-105质粒、MMP-2质粒、miR-105和MMP-2质粒。CCK-8法检测细胞增殖;Transwell法检测U251细胞侵袭;划痕实验检测细胞迁移;流式细胞术检测细胞凋亡。结果:胶质瘤组织中miR-105表达水平低于瘤旁组织,而MMP-2高于瘤旁组织(P<0.05)。MMP-2与miR-105均是脑胶质瘤术后复发的独立影响因素(P<0.05)。miR-105、MMP-2单一及联合检测预测术后复发的ROC曲线下面积(AUC)依次为0.735、0.797、0.894。miR-105可以抑制U251细胞的增殖、侵袭和迁移,并促进凋亡(P<0.05);MMP-2可以促进U251细胞的增殖、侵袭和迁移,并抑制凋亡(P<0.05)。结论:miR-105、MMP-2参与脑胶质瘤的侵袭、迁移及凋亡,二者对脑胶质瘤术后复发的预测有一定价值。

Objective: To investigate the effects of matrix metalloproteinase-2(MMP-2) and microRNA(miR)-105 on the invasion, migration and apoptosis of glioma cells, and their value in predicting postoperative recurrence. Methods: 89 cases of glioma tissues and matched paratumor tissues surgically resected in our hospital from July 2018 to July 2021 were selected. RT-PCR was used to detect the expression level of miR-105; immunohistochemical staining was used to detect the expression of MMP-2. Logistic and receiver operating characteristic (ROC) curves were used to analyze the value of MMP-2 and miR-105 in predicting postoperative recurrence of glioma. The glioma cell line U251 was selected and transfected with blank plasmids, miR-105 plasmids, MMP-2 plasmids, miR-105 and MMP-2 plasmids. Cell proliferation was detected by CCK-8 method; U251 cell invasion was detected by Transwell method; cell migration was detected by scratch assay; cell apoptosis was detected by flow cytometry. Results: The expression level of miR-105 in glioma tissues was lower than that in adjacent tissues, while MMP-2 was higher than that in adjacent tissues(P<0.05). Both MMP-2 and miR-105 were independent influencing factors of postoperative recurrence of glioma of the brain(P<0.05). The areas under the curve(AUC) of miR-105 and MMP-2 alone and in combination to predict postoperative recurrence were 0.735, 0.797 and 0.894, respectively. miR-105 could inhibit the proliferation, invasion and migration of U251 cells, and promote apoptosis(P<0.05); MMP-2 could promote the proliferation, invasion and migration of U251 cells, and inhibit apoptosis(P<0.05). Conclusion: miR-105 and MMP-2 are involved in the invasion, migration and apoptosis of glioma, and they have certain value in the prediction of postoperative recurrence of glioma.
