口腔鳞癌组织中OR2W3的表达及其临床意义

摘要:

目的:检测口腔鳞癌术后组织中嗅觉受体家族2亚家族W成员3(OR2W3)的表达,探讨其临床意义及与细胞增殖的关系。方法:收集行口腔鳞癌根治手术的患者共79例作为研究对象,选择肿瘤组织作为观察组,选择周围正常口腔黏膜组织作为对照组。留取术后的新鲜组织及石蜡包埋组织,应用免疫组化法和蛋白质印迹法检测两组中OR2W3蛋白的表达,应用实时荧光定量PCR法检测两组中OR2W3的mRNA表达,应用免疫组化法检测观察组中Ki67的表达。结果:免疫组化法检测结果:观察组中OR2W3表达的阳性率明显高于对照组,观察组中OR2W3的表达与肿物最大径、脉管累犯、颌骨累犯、淋巴结转移及TNM分期相关,而与性别、年龄及肿瘤分化程度无明显相关性。OR2W3的表达与Ki67增殖指数具有正相关性。生存分析显示,OR2W3的表达与生存时间有关。蛋白质印迹法检测结果显示,观察组中OR2W3蛋白的半定量表达明显高于对照组。实时荧光定量PCR检测结果显示,观察组中OR2W3 mRNA的表达量明显高于对照组。结论:OR2W3在口腔鳞癌组织中高表达,对病变形成和进展有重要的作用,OR2W3高表达对促进细胞增殖有明确的作用,术后检测组织中OR2W3的表达可能对判断预后有一定价值。

Objective: To detect the expression of olfactory receptor family 2 subfamily W member 3(OR2W3) in oral squamous carcinoma tissues, and to explore its clinical significance and relationship with cell proliferation. Methods: A total of 79 patients of oral squamous carcinoma were selected as the study subjects. Tumor tissues were selected as the observation group, normal oral mucosa tissues were selected as the control group. Fresh tissue and paraffin-embedded tissue were retained after operation. The expressions of OR2W3 protein were detected by immunohistochemistry and Western Blot, and the expressions of OR2W3 mRNA were detected by real-time fluorescence quantitative PCR in two groups. Ki67 was detected by immunohistochemistry in observation group. Results: Immunohistochemical results:The positive rate of OR2W3 expression was significantly higher in the observation group than that in the control group. The expression of OR2W3 was correlated with the maximum diameter of tumors, vascular recidivism, jaw recidivism, lymph node metastasis and TNM staging, but not with sex, age and differentiation in the observation group. The expression of OR2W3 was positively correlated with proliferation index. OR2W3 expression was related to survival time by survival analysis. Semi-quantitative expression of OR2W3 protein was significantly higher in the observation group than that in the control group by Western Blot methods. Expression of OR2W3 mRNA was significantly higher in the observation group than that in the control group by real-time fluorescence quantitative PCR. Conclusion: Over expression of OR2W3 plays an important role in formation and progression of oral squamous carcinoma. Higher OR2W3 expression has a definite role in promoting cell proliferation. OR2W3 expression may have a certain value in judging prognosis.
