环状RNA在肺腺癌中的差异表达分析

摘要:

目的:分析肺腺癌(LAC)组织中环状RNA (circRNA)的差异表达,探讨circRNA参与LAC发展的机制。方法:通过基因芯片筛选6例LAC患者中circRNA的差异表达谱。采用微小RNA (miRNA)靶预测软件预测circRNA和miRNA相互作用的结合位点,再分别预测最显著高表达及低表达的两条circRNA分子下游相关的靶基因,并分析和探索其中涉及的生物学通路。结果:共有19条circRNA在癌组织标本内差异表达至3倍以上,其中最显著差异上调的hsa_circRNA_100323及下调的hsa_circRNA_051620具有多个miRNA结合位点,它们可能通过各种生物学过程及通路参与LAC的发生与发展。结论:LAC患者中circRNA表达谱发生了显著变化,最显著高表达和低表达的hsa_circRNA_100323及hsa_circRNA_051620可能通过miRNA"海绵"吸附作用在LAC的发生和发展中发挥重要作用。

Objective: To analyze the differentially expressed circular RNAs(circRNAs) in patients with lung adenocarcinoma(LAC), and investigate how they regulate the development of LAC. Methods: Gene microarray was used to identify the differential expression profile of circRNAs in 6 patients with LAC. The microRNA(miRNA) target predicted software was used to predict the binding sites between circRNAs and miRNAs, and the downstream related target genes of the two circRNAs with the most significant high expression and low expression were predicted respectively, then the biological pathways involved were analyzed and explored. Results: A total of 19 circRNAs were differentially expressed at least more than 3 times in tumor tissues, the most significantly up-regulated hsa_circRNA_100323 and down-regulated hsa_circRNA_051620 had multiple binding sites of miRNAs, and they maybe involved in the occurrence and development of LAC by various biological processes and pathways. Conclusion: The circRNA expression profile in LAC has changed significantly. The most significantly up-regulated hsa_circRNA_100323 and down-regulated hsa_circRNA_051620 may play an important role in the occurrence and development of LAC through the biological function of the sponger of miRNA.
