BTG3在甲状腺乳头状癌组织中的表达及其对细胞增殖、侵袭、迁移的影响

摘要:

目的:探讨B细胞转位基因3(BTG3)在甲状腺乳头状癌组织中的表达及其对甲状腺乳头状癌TPC-1细胞增殖、侵袭、迁移的影响。方法:选择60例甲状腺乳头状癌患者的新鲜肿瘤标本及其对应的癌旁组织,采用免疫组化法测定组织中BTG3蛋白表达水平。将甲状腺乳头状癌TPC-1细胞分为BTG3过表达组、阴性对照组和空白对照组,分别于0、24、72 h加入10 μl CCK-8试剂孵育2 h,测定TPC-1细胞增殖能力(OD值),采用微孔滤膜实验测定TPC-1细胞侵袭和迁移能力,采用蛋白质印迹法测定各组TPC-1细胞的波形蛋白、N-钙粘蛋白和E-钙粘蛋白水平。结果:甲状腺乳头状癌组织中BTG3蛋白阳性率低于癌旁组织(P < 0.05)。TPC-1细胞0、24、72 h的OD值BTG3过表达组明显低于阴性对照组和空白对照组相应时间点的OD值(均P < 0.001)。TPC-1细胞BTG3过表达组细胞侵袭迁移能力和E-钙粘蛋白水平高于阴性对照组和空白对照组(均P < 0.05);波形蛋白、N-钙粘蛋白水平低于阴性对照组和空白对照组(均P < 0.05)。结论:BTG3在甲状腺乳头状癌组织中低表达;甲状腺乳头状癌细胞过表达BTG3后,其增殖、侵袭和迁移能力下降,波形蛋白、N-钙粘蛋白和E-钙粘蛋白水平发生变化。

Objective: To investigate the expression of B cell translocation gene 3 (BTG3) in papillary thyroid carcinoma and its effect on proliferation, invasion and migration of papillary thyroid carcinoma TPC-1.Methods: Sixty cases of fresh papillary thyroid carcinoma tissue and 60 corresponding paracancerous tissues were selected. The expression of BTG3 protein in the tissues was determined by immunohistochemistry.Papillary thyroid carcinoma TPC-1 cells were divided into BTG3 overexpression group, negative control group and blank control group.CCK-8 method was used to determine the proliferation (expressed as OD value) of TPC-1 cells after incubation with 10 μl CCK-8 for 0, 24 and 72 h. Transwell assay was used to determine the invasion and migration ability of TPC-1 cells. Expression levels of vimentin, N-cadherin and E-cadherin in TPC-1 cells were determined by Western blot. Results: The positive rate of BTG3 protein in thyroid papillary carcinoma was lower than that in paracancerous tissue (P < 0.05).The OD values of TPC-1 cells at the timepoint of0, 24 and 72 h in the BTG3 overexpression group were significantly lower than those of the negative control group and blank control group at the same timepoint (all P < 0.001). The invasion and migration ability and the E-cadherin protein level in BTG3 overexpression group were higher than those of the negative control group and blank control group (all P < 0.05). The levels of vimentin and N-cadherin in BTG3 overexpression group were lower than those in the negative control group and blank control group. Conclusion: BTG3 is lowly expressed in papillary thyroid carcinoma. When overexpression of BTG3 in papillary thyroid carcinoma cell, the cells proliferation, invasion and migration are decreased, and the vimentin, N-cadherin and E-cadherin protein levels change.
