未成熟同源蛋白增强基因在膀胱尿路上皮癌组织中的表达及其临床意义

摘要:

目的:观察未成熟同源蛋白增强(ERH)基因在人膀胱尿路上皮癌(BUC)组织中的表达及其临床意义。方法:从癌症基因组图谱(TCGA)数据库查到ERH基因在BUC和正常膀胱组织中存在差异性表达,采用聚合酶链式反应(qPCR)检测技术验证12对在徐州市中心医院确诊为BUC并行手术治疗的癌组织和癌旁正常膀胱组织中ERH基因的表达情况,通过BUC组织芯片和免疫组化法验证ERH基因在BUC组织中的表达,分析ERH的高表达与BUC患者的年龄、性别、肿瘤大小、T分期、淋巴结转移、临床分期等指标的关系。结果:qPCR和免疫组化结果提示,ERH基因在人BUC组织中比正常膀胱组织表达增多,差异有统计学意义(P < 0.05)。组织芯片的免疫组化检测结果也显示,ERH基因在BUC组织中表达明显高于正常膀胱组织(P < 0.05),但ERH表达量与肿瘤的T分期、临床分期、转移、复发等指标均无相关性(均P > 0.05)。结论:ERH基因在BUC中呈高表达,表达量与临床肿瘤学特征无相关性。

Objective: To observe the expression of enhancer of rudimentary homolog (ERH) gene in human bladder urothelial carcinoma (BUC) tissues and its clinical significance. Methods: From the Cancer Genome Atlas (TCGA) database, the differential expression of ERH gene in BUC and normal bladder tissues were detected. Polymerase chain reaction (qPCR) was used to verify the expression of ERH gene in 12 pairs of BUC tissues and normal bladder tissues from BUC patients diagnosed in Xuzhou Central Hospital. The expression of ERH gene in the cancer tissues and adjacent normal bladder tissues were verified by BUC tissue microarray and immunohistochemistry. The relationship between the high expression of ERH and the age, sex, tumor size, T stage, lymph node metastasis, clinical stage and other indicators of the BUC patients were analyzed. Results: The results of qPCR and immunohistochemistry showed that the expression of ERH gene in human BUC tissues were higher than those in normal bladder tissues, and the difference were statistically significant (P < 0.05). The immunohistochemistry results of tissue microarray also showed that the expression of ERH gene in BUC tissues was significantly higher than that in normal bladder tissues (P < 0.05). Moreover, there was no correlation between ERH expression and tumor T stage, clinical stage, metastasis, recurrence and so on (P > 0.05). Conclusion: The ERH gene is highly expressed in BUC, but the expression level is not correlated with clinical oncology.
