结肠腺癌中eIF3d和eIF3e表达的研究

摘要:

目的:检测结肠腺癌组织中真核翻译起始因子3d(eIF3d)和真核翻译起始因子3e(eIF3e)的表达,分析其相关性及对预后判断的价值。方法:选择67例结肠腺癌患者作为观察组,留取术后石蜡包埋组织及新鲜组织,以34例距肿物边缘>5 cm的正常结肠黏膜组织作为对照组。应用免疫组化法检测两组石蜡包埋组织中eIF3d和eIF3e蛋白的表达,应用蛋白质印迹法检测两组新鲜标本中eIF3d和eIF3e蛋白的半定量表达,应用荧光PCR技术检测两组新鲜标本中eIF3d和eIF3e mRNA的表达。结果:免疫组化结果显示,两组中eIF3d和eIF3e蛋白表达阳性率差异有统计学意义(P<0.05)。观察组中eIF3d和eIF3e表达均与病变的增殖指数、肿瘤最大径、脉管累犯和淋巴结转移密切相关(P<0.05),eIF3e的表达还与肿瘤浸润深度密切相关(P<0.05)。相关分析显示,观察组中eIF3d和eIF3e的表达呈正相关(r=0.52,P=0.027 4)。生存分析显示,观察组中eIF3d和eIF3e的阳性率与生存时间相关(P<0.05)。蛋白质印迹法结果显示,两组中eIF3d和eIF3e半定量表达的差异有统计学意义(P<0.05);荧光PCR结果显示,两组中eIF3d和eIF3e mRNA的表达差异有统计学意义(P<0.05)。结论:eIF3d和eIF3e蛋白及mRNA在结肠腺癌组织中的表达升高,在肿瘤的形成和发展中具有促进作用。eIF3d与eIF3e的表达呈正相关,联合检测组织中eIF3d和eIF3e的表达对预后判断有一定价值。

Objective: To detect the expressions of eukaryotic translation initiation factor 3d(eIF3d) and eukaryotic translation initiation factor 3e(eIF3e) in colorectal adenocarcinoma, analyze their relationship and prognostic value. Methods: 67 cases of colorectal adenocarcinoma were chosen as observation group, 34 normal colon mucosa tissue as control group. Expressions of eIF3d and eIF3e were detected by IHC methods in paraffin wax specimens in two groups. Expressions of eIF3d and eIF3e were detected by Western blotting in fresh tissue in two groups. Expressions of eIF3d and eIF3e mRNA were detected by qPCR in fresh tissue in two groups. Results: The positive rate of eIF3d and eIF3e was obvious different in two groups(P<0.05). Expressions of eIF3d and eIF3e were closely related to proliferation index, maximum diameter, vascular involvement and lymph node metastasis(P<0.05). Expression of eIF3e was also closely related to invasive depth(P<0.05). There was positive correlation between eIF3d and eIF3e expression in observation group(r=0.52, P=0.027 4). Expressions of eIF3d and eIF3e were closely related to survival time in observation group(P<0.05). Expressions of eIF3d and eIF3e mRNA was obvious different in two groups(P<0.05). Conclusion: Higher protein and mRNA expressions of eIF3d and eIF3e may promote the occurrence and development in colorectal adenocarcinoma. Expression of eIF3d and eIF3e is positively correlated. Combined detection of eIF3d and eIF3e has a certain significance in judging prognosis.
