Objective:To assess whether the levels of prokineticin 1(PROK) in cumulus cells derived from patients undergoing invitro fertilization-embryo transfer(IVF-ET) were related to the embryo quality and clinical outcomes in IVF cycle. Methods:A total of one hundred and sixty-two IVF-ET patients were enrolled in this study, and the protein levels of PROK1 in cumulus cells were measured by Western blot. The patients were divided into two groups according to whether the PROK1 protein levels in their cumulus cells(CCs) were above or below the mean level detected in all the patients. The difference of clinical data, embryo quality and pregnancy outcome, as well as p53, cleaved-caspase-3 and PCK1 protein levels, between the two groups was compared. Moreover, relationship between PROK1 levels and glucose metabolism in CCs was also compared. Results:The basal follicle-stimulating hormone(FSH) level, retrieved oocyte, and the rate of maturiration,fertilization, cleavage, available embryo, high quality embryos,implantation and pregnancy were significantly higher in high PROK1 group than those in low PROK1 group, the differences between the two groups being statistically significant(all P<0.05). Moreover, the expression of cleaved-caspase-3, PCK1 protein and glucose content was also higher in high PROK1 group than that in low PROK1 group with statistically significant difference(all P<0.05). Correlation analysis demonstrated that cleaved-caspase-3, PCK1 protein levels and glucose contents were negatively correlated with PROK1 protein(P<0.05). Conclusion:The present findings imply that the PROK1 level in CCs is associated with embryo quality and pregnancy outcome of IVF-ET patients. Over-expression of PROK1 may have positive effects on oocyte maturation and embryo development. |
