补气增液汤对慢传输型便秘大鼠肠道功能、肠神经递质及Cajal间质细胞的影响

摘要:

目的:探究补气增液汤对慢传输型便秘(STC)大鼠排便量、肠道功能、肠神经递质、Cajal间质细胞(ICC)及干细胞因子(SCF)/干细胞生长因子受体(c-kit)信号通路的影响。方法:选取50只雄性SD大鼠随机分为正常组、模型组、低剂量补气增液汤组、高剂量补气增液汤组和莫沙必利组,每组10只;采用复方地芬诺酯建立STC大鼠模型,检测造模前、造模期、治疗期大鼠粪便数量及含水率;检测各组大鼠肠神经递质一氧化氮(NO)、一氧化氮合酶(NOS)、P物质(SP)、血管活性肠肽(VIP)含量以及c-kit、SCF蛋白的表达。结果:与模型组相比,补气增液汤能够增加STC大鼠粪便数量及粪便含水率(P<0.05);与正常组比较,模型组大鼠结肠SP、VIP、NO及NOS含量升高(P<0.05);而与模型组比较,补气增液汤组神经递质VIP、NO和NOS水平下降(P<0.05),c-kit、SCF蛋白表达水平提高(P<0.05)。结论:补气增液汤能够改善便秘症状,其机制可能与减少结肠NO、NOS、VIP含量及调控SCF/c-kit信号通路,进而增强肠道平滑肌运动有关。

Objective:To explore the effects of Buqizengye decoction on defecation, intestinal function, intestinal neurotransmitters, Cajal interstitial cells(ICC) and SCF/c-kit signaling pathway in rats with slow transit constipation(STC). Methods:Fifty male SD rats were randomly divided into normal group, model group, low-dose group of Buqizengye decoction, high-dose group of Buqizengye decoction and Mosapride group with 10 rats in each group. The rat model of STC was established to detect the amount of feces and water content in rats before, during and after treatment.The intestinal neurotransmitters NO, NOS, SP and VIP, and the expression of c-kit and SCF protein were detected in each group. Results:Compared with the model group, Buqi zengye decoction could increase the fecal amount and water content of STC rats(P<0.05). Compared with the normal group, the levels of SP, VIP, NO and NOS in the colon of the model group increased(P<0.05). Compared with the model group, the levels of VIP, NO and NOS decreased in the Buqizengye decoction group(P<0.05), while the expression of c-kit and SCF protein was increased in the Buqizengye decoction group(P<0.05). Conclusion:Buqizengye decoction can improve the symptoms of constipation, and its mechanism may be related to the reduction of the content of NO, NOS, VIP in colon and the regulation SCF/c-kit signaling pathway, which can improve the smooth muscle movement of the intestine.
