槐耳清膏对乳腺癌MCF-7细胞裸鼠移植瘤的抑制作用及其与调控miR-126表达的关系

摘要:

目的:探讨槐耳清膏对乳腺癌MCF-7细胞裸鼠致瘤性生长抑制作用及其与调控miR-126表达的关系。方法:建立裸鼠乳腺癌MCF-7细胞皮下移植瘤模型,随机将裸鼠分为两组,实验组给予槐耳清膏(浓度150 mg·ml-1)0.2 ml灌胃,对照组采用0.9%生理盐水0.2 ml灌胃,每天两次,共30 d,观察裸鼠瘤体积及瘤体重的变化;RT-PCR检测裸鼠移植瘤组织中miR-126的表达。结果:(1)裸鼠在接种MCF-7细胞3周后,实验组裸鼠移植瘤体重为(0.20±0.07)g,而对照组裸鼠移植瘤体重为(0.57±0.32)g,实验组裸鼠移植瘤体重明显低于对照组,差异具有统计学意义(P<0.01)。(2)实验组裸鼠移植瘤体积为(0.32±0.74)cm3,而对照组裸鼠移植瘤体积为(0.84±0.62)cm3,实验组移植瘤体积较对照组小,差异具有统计学意义(P<0.01)。(3)裸鼠移植瘤组织中miR-126表达实验组是对照组的2.406倍,差异具有统计学意义(P<0.05)。结论:槐耳清膏能抑制裸鼠乳腺癌MCF-7细胞的生长,其机制可能是通过上调miR-126的表达从而抑制乳腺癌的发生和发展。

Objective:To explore the inhibitive effect of huaier cream on xenografted human MCF-7 cells in nude mice and its role in the regulation of miR-126 expression. Methods:Tumor models were established by subcutaneously transplanting human MCF-7 cells into nude mice and then which were randomly divided into two groups. The experimental group received 0.2 ml Huaier cream (150 mg·ml-1) lavage twice per day for a period of 30 days, while 0.2 ml of 0.9% saline was used in the control group. After sacrificing the mice, tumor volume and weight were accordingly measured and recorded. RT-PCR was used to measure the expression of miR-126. Results:(1) All nude mice developed tumors at the transplanted site 3 weeks after inoculation with MCF-7 cells. The transplanted tumors were significantly lighter in the experimental group[(0.20±0.07)] g when compared with those in the control group[(0.57±0.32)] g (P<0.01). (2) Tumor volumes were significantly lower in the experimental group than those in the control group[(0.32±0.74) cm3 vs (0.84±0.62) cm3, P<0.01]. (3) The expression of miR-126 in the experimental group was 2.406 times as high as in the control group, the difference was statistically significant (P<0.05). Conclusion:Huaier cream can significantly inhibit the growth of MCF-7 cells in nude mice,which might be through the upregulation of miR-126 expression, thereby inhibiting the occurrence and development of breast cancer.
