蒲公英甾醇对脂多糖诱导乳腺炎大鼠的抗炎作用及其机制

摘要:

目的:探讨蒲公英甾醇对脂多糖(LPS)诱导乳腺炎大鼠的抗炎作用及其相关机制。方法:50只产后大鼠随机分为对照组、模型组、阳性对照组、蒲公英甾醇低剂量组和蒲公英甾醇高剂量组,乳导管内灌注LPS建立乳腺炎大鼠模型,分别以不同剂量蒲公英甾醇(5、10 mg·kg-1)及地塞米松(5 mg·kg-1)给药,24 h后取乳腺组织。进行组织切片观察乳腺组织的病理变化,采用ELISA法检测TNF-α、IL-1β和IL-6水平,采用RT-PCR检测乳腺组织iNOS、COX-2 mRNA表达水平,免疫印迹法检测乳腺组织NF-кB、MAPKS信号通道蛋白水平。结果:与模型组比较,蒲公英甾醇低剂量组和高剂量组大鼠乳腺组织TNF-α、IL-1β、IL-6、iNOS mRNA、COX-2 mRNA、磷酸化I-κB、NF-κB-p65、JNK、ERK、p38的表达水平显著下降(P<0.05)。结论:蒲公英甾醇可降低iNOS和COX-2 mRNA表达,抑制NF-κB、MAPKs信号通路和促炎细胞因子的产生,对乳腺炎发挥抗炎作用。

Objective:To investigate the anti-inflammatory effect and related mechanisms of taraxasterol on lipopolysaccharide (LPS)-induced mastitis rats. Methods:Fifty rats were randomly divided into normal group, model group, positive control group, taraxasterol low dose group and taraxasterol high group. Mastitis model rats were establish by LPS injected into mammary ducts. Mammary tissue was obtained after 24 h since administration of different doses of taraxasterol (5 mg·kg-1, 10 mg·kg-1) or dexamethasone(5 mg·kg-1). The pathological changes of breast tissue were observed by tissue section. ELISA was used to determine the levels of TNF-α, IL-1β and IL-6. RT-PCR was applied to determine the expression of iNOS、COX-2 mRNA in mammary tissue. Western-blot was used to determine the protein levels of NF-кB, MAPKS signal pathways in mammary tissue. Results:Compared with model group, expression levels of TNF-α, IL-1β, IL-6, iNOS mRNA, COX-2 mRNA, phosphorylate I-κB, NF-κB-p65, JNK, ERK, p38 in taraxasterol low dose group and taraxasterol high group were significantly decreased(P<0.05). Conclusion:Taraxasterol plays anti-inflammatory effect by reducing the expression of iNOS and COX-2 mRNA, inhibiting NF-κB and MAPKs signal pathways, inhibiting the production of inflammatory cytokines.
