PPARγ激动剂罗格列酮对胰岛β细胞中CASK基因表达的影响

摘要:

目的:探讨过氧化物酶体增殖剂激活受体(PPAR)γ激动剂罗格列酮对钙/钙调蛋白依赖性丝氨酸蛋白激酶(CASK)的影响,探索CASK是否参与罗格列酮对胰岛素分泌的调控作用。方法:用不同浓度罗格列酮处理胰岛β细胞系INS-1细胞检测氯化钾刺激胰岛素分泌(KSIS);用不同浓度及时间罗格列酮处理INS-1细胞,检测CASK的mRNA、蛋白及定位变化;观察干扰CASK后罗格列酮对KSIS的影响情况。结果:罗格列酮处理后INS-1细胞的KSIS功能明显增强(P<0.05);不同浓度及时间罗格列酮处理INS-1细胞后CASK的mRNA和蛋白水平呈剂量及时间依赖性增加(P<0.05);干扰CASK后罗格列酮对KSIS的影响差异无统计学意义(P>0.05)。结论:PPARγ激动剂罗格列酮上调INS-1细胞CASK基因的表达,但CASK并不参与罗格列酮对胰岛分泌的调控作用。
