Objective:To investigate the effects and mechanisms of α-lipoic acid (ALA) in atherosclerosis (AS) rabbits. Methods:AS lesions were induced in rabbits receiving liquid nitrogen treatment and high fat diet. According to different treatment, rabbits were divided into normal group, model group and model+ALA group. Pathological conditions of arterial tissues and the changes of lipids were detected. The expression of cell adhesion molecules (ICAM-1, VACM-1, P-selection) and NF-κB protein were measured in arterial tissues. Results:Compared with the normal group, the thickness of arterial intima and the levels of serum lipids increased and plaques were observed in the model group, meanwhile, the expression of cell adhesion molecules (ICAM-1, VACM-1, P-selection) and NF-κB in arterial tissues significantly increased in the model group. Compared with the model group, the area of plaques and the levels of serum lipids reduced in the model+ALA group; and simultaneously the expression of cell adhesion molecules and NF-κB also decreased in the model+ALA group. Conclusion:α-lipoic acid may inhibit the expression of cell adhesion molecules by suppressing the expression of NF-κB, leading to the inhibition of vascular inflammation, which may be one of the mechanisms of ALA on improving pathology of AS.
