宫颈癌患者血浆miRNA-214的表达水平及其对宫颈癌细胞的影响

摘要:

目的:探讨微小RNA-214(miRNA-214)在宫颈癌患者血浆中表达水平的差异及其对宫颈癌细胞增殖、细胞周期和凋亡的影响。方法:将2016年1月至2018年1月在中国科学院合肥肿瘤医院就诊、经宫颈活检病理确诊为宫颈浸润癌的50例患者作为宫颈癌组,同时选取相同年龄段体检健康者50例作为健康对照组。采用实时荧光定量PCR法对两组血浆中的miRNA-214进行检测并比较。采用脂质体介导的转染方法将miRNA-214模拟物转染,CCK-8法检测宫颈癌细胞SiHa的增殖、细胞周期和凋亡。结果:血浆中miRNA-214的表达宫颈癌组与健康对照组比较,差异有统计学意义(P<0.05)。用CCK-8法检测SiHa发现,miRNA-214模拟物转染组细胞的生长速度明显慢于对照组,即miRNA-214对细胞生长速度有抑制作用,差异有统计学意义(P<0.05)。与宫颈癌对照组[转染miRNA-阴性(即miRNA-NC)]相比,宫颈癌转染miRNA-214组(脂质体转染miRNA-214模拟物)中,宫颈癌S期细胞比例明显降低,凋亡细胞比例明显升高(P<0.05),G2/M期细胞比例明显降低(P<0.05)。结论:宫颈癌患者血浆中miRNA-214表达下调,作为宫颈癌早期诊断分子标志物有一定准确性。miRNA-214可以抑制宫颈癌细胞SiHa进入S期,并且促进SiHa细胞凋亡,miRNA-214可能在宫颈癌的发生与发展中发挥着抑癌作用。
