Objective:To observe the effect of calcitriol on serum inflammatory cytokines in Hashimoto's thyroiditis rats and to explore its clinical significance.Methods:36 rats were randomly divided into control group, model group and calcitriol group, each group had 12 rats. Rats in model group and calcitriol group were made thyroiditis model by high iodine. Rats in calcitriol group were treated with calcitriol. The morphological changes of thyroid tissue were observed in each group.Thyroglobulin antibody (TGAb), anti-thyroid peroxidase autoantibody (TPOAb), tumor necrosis factor-α (TNF-α),interleukin-1 (IL-1) and interleukin-6 (IL-6) were measured by enzyme-linked immunosorbent assay (ELISA).The levels of free triiodothyronine (FT3), free thyroxine (FT4) and thyroid stimulating hormone (TSH) were measured by chemiluminescence method.Results:In the control group, thyroid follicles were oval or round,the thyroid epithelial cells arranged evenly and neat,had no plasma cells and lymphocyte infiltration.In the model group, the thyroid follicle was arrangement disorder,and had plasma cell infiltration, thyroid epithelial cells arranged disorder, had significant lymphocyte infiltration.In the calcitriol group,thyroid follicle arrangement was basic rules,had occasional plasma cell infiltration,thyroid epithelial cells can be arranged,had a small amount of lymphocyte infiltration.The levels of TGAb, TPOAb, FT3 and FT4 in the model group and the calcitriol group were higher than those in the control group (P<0.05),the levels of TSH were lower than that in the control group (P<0.05).The levels of TGAb and TPOAb in the calcitriol group were lower than those in the model group (P<0.05).There was no significant difference in FT3, FT4, TSH between the calcitriolol group and the model group (P>0.05).The levels of serum TNF-α, IL-1 and IL-6 in the model group and the calcitriol group were higher than those in the control group (P<0.05).The levels of serum TNF-α, IL-1 and IL-6 in the calcitriol group were lower than those in the model group (P<0.05).Conclusion:Calcitriol can exert therapeutic effect on Hashimoto's thyroiditis rats by lowering serum levels of TNF-α, IL-1 and IL-6.[Key words] calcitriol; Hashimoto's thyroiditis; rats; tumor necrosis factor-α; interleukin-1; interleukin-6 |
