骨髓间充质干细胞培养上清液在未成熟鼠脑损伤中的免疫调节作用

摘要:

目的:观察骨髓间充质干细胞上清液对未成熟鼠脑损伤模型炎症因子白细胞介素-6(IL-6)及转化生长因子-β(TGF-β)的影响,探讨骨髓间充质干细胞旁分泌作用干预未成熟鼠脑损伤的机制。方法:采用健康SD大鼠的24只新生3 d乳鼠建立早产儿脑损伤模型,分为对照组、磷酸缓冲溶液(PBS)干预组和细胞培养上清液干预组。模型制作成功后,各组侧脑室立体定向移植PBS及上清液,制备侧脑室HE染色病理切片,采用酶联免疫吸附测定法检测各组1、3、5 d IL-6及TGF-β的分泌趋势及分泌浓度。结果:早产儿脑损伤模型分泌IL-6随天数递增而增多,上清液干预组分泌IL-6的量明显少于对照组及PBS干预组,分泌TGF-β的量明显高于对照组及PBS干预组。结论:骨髓间充质干细胞上清液可以改善未成熟鼠脑损伤的预后,降低促炎因子,增加保护性因子的分泌量。

Objective:To observe the impacts of culture supernatant of bone marrow mesenchymal stem cells(BMSCs) on inflammatory factor interleukin-6(IL-6), transforming growth factor-β(TGF-β) in immature rat brain injury model and investigate the mechanism of BMSCs paracrine interfering in immature rat brain injury. Methods:24 healthy three-day-old Sprague Dawley(SD) rats were selected and immature rat brain injury model was established. These rats were randomly divided into control group, PBS group and supernatant group. The supernatant group were given culture supernatant of BMSCs through intracerebroventricular injection. The PBS group received PBS and the control group received nothing. The secretion level of IL-6 and TGF-β at 1, 3, 5 d in each group were detected by HE staining and ELISA. Results:The secretion of IL-6 in immature rat brain injury model increases with days. The secretion of IL-6 in supernatant group was evidently less than control group and PBS group while the secretion of TGF-β was much more higher than the other two groups. Conclusion:Supernatant of BMSCs can improve the prognosis of immature rat brain injury, increase the secretion of protective factors and decrease the proinflammatory factor.
