血栓素A2及PI3K/AKT信号通路在早发型子痫前期的作用研究

摘要:

目的:研究血栓素A2(TXA2)及磷脂酰肌醇3-激酶(PI3K)/蛋白激酶B(AKT)信号通路在早发型子痫前期的作用和意义。方法:选取本院2015年5月至2017年5月收治的60例子痫前期孕妇为观察组,同期30例正常孕妇为对照组,采用免疫组织化学法检测两组胎盘组织中TXA2的表达,采用Caspase 3活性检测试剂盒检测凋亡蛋白Caspase 3表达,采用Real-time PCR和Western blotting检测患者血清和胎盘组织中PI3K、AKT的基因和磷酸化蛋白的表达。结果:与对照组相比,观察组患者血清TXA2、PI3K、AKT含量明显升高,Caspase 3活性明显升高,差异具有统计学意义(P<0.05);观察组患者胎盘组织内TXA2、PI3K和AKT的蛋白和mRNA表达显著升高,差异具有统计学意义(P<0.05)。与对照组相比,观察组患者胎盘组织内TXA2蛋白平均光密度水平明显升高,差异具有统计学意义(P<0.05)。结论:早发型子痫前期患者体内TXA2呈异常过表达状态,且PI3K/AKT信号通路在早发型子痫前期的发病过程中具有重要的作用。

Objective:To study the role and significance of thromboxane A2 (TXA2) and PI3K/AKT signaling pathway in early preeclampsia. Methods:Sixty cases of pregnant women with preeclampsia were selected as the observation group from May 2015 to May 2017. 30 cases of normal pregnant women were selected as the control group. The expression of thromboxane A2 in placenta tissue was detected by immunohistochemistry.The expression of Caspase 3 was detected by Caspase 3 activity assay kit. Real-time PCR and Western blotting were used to detect the expression of PI3K, AKT and phosphorylated protein in serum and placenta of two groups. Results:Compared with the control group, the contents of TXA2, PI3K and AKT in the blood of the observation group were significantly increased (P<0.05), the activity of Caspase 3 was significantly increased (P<0.05).The expression of TXA2, PI3K and AKT in the placenta of the observation group were significantly increased compared with those in the control group (P<0.05).Compared with the control group, the average optical density of TXA2 protein in the placenta of the observation group was significantly increased (P<0.05). Conclusion:Thromboxane A2 in early-onset preeclampsia patients is abnormally overexpressed, and PI3K/AKT signaling pathway plays an important role in the pathogenesis of early preeclampsia.
