槐耳清膏对人乳腺癌MCF-7及MDA-MB-231细胞增殖的影响

摘要:

目的:研究槐耳清膏对人乳腺癌MCF-7及MDA-MB-231细胞增殖的影响。方法:配制不同浓度的槐耳清膏(3、6、9 mg·ml-1)作用于MCF-7及MDA-MB-231细胞48 h后,MTT比色法测定槐耳清膏各浓度组对MCF-7及MDA-MB-231细胞的增殖抑制率。结果:槐耳清膏作用于乳腺癌MCF-7细胞48 h后,与阴性对照组比较,各浓度组的OD值逐渐下降,在槐耳清膏浓度为3、6、9 mg·ml-1时其细胞抑制率分别为(17.4±1.4)%、(58.6±3.7)%、(76.1±2.9)%,差异具有统计学意义(P<0.05)。槐耳清膏作用于乳腺癌MDA-MB-231细胞48 h后,与阴性对照组比较,各浓度组的OD值变化不大,在槐耳清膏浓度为3、6、9 mg·ml-1时其细胞抑制率分别为(10.3±1.7)%、(15.1±2.6)%、(17.8±3.9)%,差异无统计学意义(P>0.05)。结论:槐耳清膏对人乳腺癌MCF-7细胞的增殖具有明显的抑制作用,随着药物浓度的增加抑制作用更加明显。槐耳清膏对乳腺癌MDA-MB-231细胞无明显的抑制增殖作用。

Objective:To investigate the effects of Huaier on proliferation of human breast cancer MCF-7 and MDA-MB-231 cells. Methods:MCF-7 and MDA-MB-231 cells at the logarithmic phase were treated with 3, 6 and 9 mg·ml-1 Huaier cream for 48 h, respectively. MTT was used to determine OD value, and the inhibitory rate was calculated. Results:Compared with the negative control group, OD values in MCF-7 cells of each concentration group was gradually decreased, the inhibitory rates of MCF-7 cells were (17.4±1.4)%, (58.6±3.7)% and (76.1±2.9)%, when the concentrations of Huaier cream were 3, 6, and 9 mg·ml-1, respectively, with statistically significant difference (P<0.05). Compared with the negative control group, OD values were not obviously changed in human breast cancer MDA-MB-231 cells after treatment with Huaier cream for 48 h and the inhibitory rates of MDA-MB-231 cells were (10.3±1.7)%, (15.1±2.6)% and (17.8±3.9)%, when the concentrations of Huaier cream were 3, 6, and 9 mg·ml-1, respectively, and there was no statistically significant difference (P>0.05). Conclusion:Huaier cream can obvious inhibit the proliferation of human breast cancer line MCF-7, the inhibitory role is enhanced by increasing the drug's concentration. While it does not inhibit the proliferation of human breast cancer cell line MDA-MB-231 obviously.
