AEG-1调控eIF4E表达促进胃癌细胞迁移的作用机制

摘要:

目的:探讨AEG-1调控eIF4E表达促进胃癌细胞迁移的作用机制。方法:在SGC7901胃癌细胞株上用质粒转染或siRNA转染的方法过表达或沉默AEG-1的表达,用Western blotting法检测eIF4E及上皮-间质样转化(EMT)的标志物表达的变化,用transwell实验检测胃癌细胞的迁移。结果:过表达AEG-1诱导SGC7901细胞发生EMT,同时上调eIF4E的表达;沉默AEG-1的表达抑制细胞发生EMT,同时下调eIF4E的表达;沉默AEG-1的表达抑制CXCL12诱导的细胞迁移,同时下调eIF4E的表达;过表达eIF4E部分逆转由于沉默AEG-1的表达引起的胃癌细胞迁移的抑制作用。结论:AEG-1通过上调eIF4E表达促进胃癌细胞的迁移。

Objective:To explore the effect and mechanism of AEG-1 in the metastasis of gastric cancer via the regulation of eIF4E. Methods:Overexpress or silence the expression of AEG-1 by plasmid transfection or siRNA transfection on SGC7901 gastric cancer cell lines. Western blotting analysis was used to detect the protein level of eIF4E and epithelial-mesenchymal transition (EMT). Cell migration was detected by transwell assay. Results:Overexpression of AEG-1 induced EMT as well as upregulation of eIF4E expression. Silence of AEG-1 expression inhibited EMT in parallel with downregulation of eIF4E expression. Silence of AEG-1 expression inhibited CXCL12-induced cell migration in parallel with downregulation of eIF4E expression. Overexpression of eIF4E promoted cell migration in AEG-1-silencing induced inhibition of cell migration. Conclusion:AEG-1 induces migration of gastric cancer cell via upregulation of eIF4E expression.
