IL-11Rα介导的分子探针在宫颈癌模型中的显像研究

摘要:

目的:探讨IL-11Rα介导的核素标记探针99Tcm-DTPA-c(CGRRAGGSC)在宫颈癌模型中的显像特性。方法:采用Western Blot法检测宫颈癌Hela细胞株IL-11Rα表达水平,免疫荧光进行HELA细胞体外结合实验;然后合成核素标记探针,对其标记率及稳定性进行检测,最后进行荷瘤裸鼠模型SPECT显像。结果:IL-11Rα在HELA细胞中呈明显高表达;合成探针即刻标记率达96.62%,放置于室温和人新鲜血清孵育12 h后放射化学纯度仍在90%;通过瘤体间质注射99Tcm-DTPA-c(CGRRAGGSC)见肿瘤局部呈明显的放射性浓聚且持续较长时间,而游离99TcmO4-注射于瘤体内30 min即见肿瘤浓聚部位浅淡。结论:标记探针可由IL-11Rα介导对宫颈癌进行特异靶向性显像,有望为高表达IL-11Rα的恶性肿瘤提供早期、特异诊断及靶向治疗。

Objective: To explore the imaging identity of nuclide labelled probe99Tcm-DTPA-c (CGRRAGGSC)in cervical cancer model in vivo. Methods: Western Blot method was used to detect the expression of IL-11Rα in cervical cancer HELA cells, and HELA cells binding assay in vitro was conducted with immunofluorescence. The labeling rate and stability were detected, SPECT imaging was made in nude mice bearing tumor. Results: IL-11Rα was highly expressed in HELA cells. The instant labeling rate of the synthetic probe was 96.62%, and the radiochemical purity could be kept at 90% when kept it at room temperature and fresh human serum for 12 h. The interstitial injection of 99Tcm-DTPA-c(CGRRAGGSC)showed a significant concentration of radioactivity and lasted for a long time, while the free99TcmO4- was injected into the tumor,it showed that the tumor concentrated was weak after 30 min. Conclusion: The marker probes can be used to carry out specific targeted imaging of cervical cancer by IL-11R alpha, which is expected to provide early diagnosis and targeted therapy for malignant tumors with high expression of IL-11R alpha.
