Objective:To observe the protective effect of tanshinone ⅡA on testicular torsion and return to testicular injury in adult rats and to explore its mechanism. Methods:36 male SD rats aged 7 weeks were randomly divided into 3 groups:sham operation group, saline group and tanshinone ⅡA group, 12 rats in each group. The rats were sacrificed at 72 hours and 4 weeks after operation. Six rats in each group and left testes were subjected to HE staining and histological injury score, Tunel staining, and Bcl-2 protein test (immunoblotting), reactive oxygen (ROS), superoxide dismutase (SOD) and malondialdehyde (MDA) were measured. Serum testosterone levels were measured as well. Results:Compared with the saline group, the testis tissue injury was significantly reduced in the tanshinone ⅡA group, the apoptotic cells were decreased and the expression of Bcl-2 was increased. The levels of SOD and the level of serum testosterone in rats was significantly higher than that in saline group (P<0.05). Conclusion:Single intraperitoneal injection of tanshinone Ⅱ has protective effect on unilateral testicular torsion injury in adult rats, possibly through the anti-apoptotic effect in early stage, and may play an antioxidant role in the long term. |
