蛋白赖氨酸甲基转移酶SET7/9表达对肝癌细胞增殖、凋亡和迁移的影响

摘要:

目的:探讨SET7/9表达对肝癌细胞增殖、凋亡和迁移的影响。方法:对SMMC-7721与Bel-7404肝癌细胞株分别转染SET7/9表达干扰质粒(shRNA-SET7/9)、SET7/9表达质粒(pSET7/9),采用CCK8法检测转染后24、48、72、96 h的OD值;用流式细胞术检测转染48 h的凋亡率;用Transwell实验测定细胞迁移力。结果:与对照组比较,SMMC-7721细胞转染shRNA-SET7/9后OD值呈时间依赖性下降,凋亡率增高[(26.8±3.4)%vs(11.4±3.9)%],迁移力明显降低(21.7±7.8 vs 104.8±11.3);而Bel-7404细胞转染pSET7/9后OD值呈时间依赖性增加,凋亡率下降[(8.7±3.6)%vs(16.4±3.1)%],迁移力增强(142.5±11.1 vs 104.7±10.5)。结论:肝癌细胞中SET7/9表达呈现肿瘤促进因子效应,相关机制值得深入研究。

Objective:To investigate the effect of SET7/9 expression on proliferation, apoptosis and migration in liver cancer cells. Methods:Liver cancer cell lines SMMC-7721 and Bel-7404 were transfected with shRNA-SET7/9 and pSET7/9 to reduce or induce SET7/9 expression, respectively. The cell viability was determined by CCK8 assay and OD value was measured 24, 48, 72 and 96 h after transfection. Apoptosis was determined 48 h after transfection by using flow cytometry. Migratory response of the transfected cells was examined by Transwell migration assay. Results:Compared with the control groups, cell viabilities declined time-dependently and apoptosis rate[(26.8±3.4)% vs(11.4±3.9)%] increased in SMMC-7721 cells with knockdown of SET7/9, but time-dependent raise of cell viabilities and decreased apoptosis rate[(8.7±3.6)% vs(16.4±3.1)%] were found in Bel-7404 cells with over-expression of SET7/9. The ability of migration was obviously inhibited(21.7±7.8 vs 104.8±11.3)in SET7/9 knockdown SMMC-7721 cells and enhanced(142.5±11.1 vs 104.7±10.5)in SET7/9 over-expression Bel-7404 cells. Conclusion:Our data suggest that SET7/9 functions as an tumor promoting factor in hepatic cancer cells and the related mechanism is worth further studying.
