Objective:To optimize and evaluate the cerebral ischemia reperfusion model of mice by means of ligation of internal carotid. Methods:Ninety healthy male Kunming mice were randomly divided into 3 groups with 30 in each group,i.e., sham operation group(Sham), ligation of internal carotid artery(LICA)group and ligation of external carotid artery(LECA)group. Assessment for neurological severity scores(NSS)was conducted to evaluate the success rate of the model and 2,3,5-triphenyhe trazolium chloride(TTC)staining was used to measure the volume ratio of cerebral infarct 24 h after operation.The correlation between the volume ratio of cerebral infarct and NSS was evaluated. The mice weight was measured and Clark's general deficits scoring was performed to evaluate the mice general survival condition 24 h,72 h,1 w after operation, respectively. Results:TTC staining confirmed that LICA method could induce evident focal cerebral ischemia, and the success rate of model reached 93.3% 24 h after operation. Compared with the LECA method, there was a better coherence between NSS and TTC staining in LICA group, as well as a significant correlation between the volume ratio of cerebral infarct measured by TTC staining and NSS. The mortality of mice 24 h, 72 h and 1 w after operation was lowered by the use of LICA surgical method and the rate of change in body weight of mice was reduced, so was the Clark's general deficit score. Conclusion:In the cerebral ischemia reperfusion model of mice, the application of LICA surgical method can improve the success rate of model and the survival rate and general functional status of mice, and increase the correlation between size of infarct focus and NSS.
