介导细胞凋亡的caspase-3在卵巢癌中的表达和放疗敏感性的关联性研究

摘要:

目的:研究介导细胞凋亡的caspase-3在卵巢癌中的表达和放疗敏感性的关系。方法:通过对临床不同时期卵巢组织样本的收集及凋亡相关因子mRNA和蛋白的检测,以及免疫组化法定位、定量caspase-3在不同时期卵巢癌中的变化来研究不同时期卵巢癌细胞凋亡情况。通过射线照射细胞然后对细胞进行计数统计,以研究不同种类卵巢癌细胞的放射敏感性。结果:免疫组化、PCR、Western Blot结果显示,随着卵巢细胞的癌变,凋亡的存活基因bcl2被激活,凋亡致死基因bax被抑制,凋亡执行蛋白caspase-3数量下降。同时射线照射和细胞统计结果显示,不同类型卵巢癌细胞对放射敏感性不同。结论:caspase-3蛋白数量随着卵巢癌变而下降,而对于卵巢癌变细胞来说,caspase-3蛋白含量越多,放射敏感性越强。

Objective: To study the caspase-3 mediated apoptosis in ovarian cancer, and the relationship between the caspase-3 expression and radiation sensitivity. Methods: Through the clinical ovarian tissue samples collected at different periods and apoptosis related gene mRNA and protein detection, and immunohistochemical legal, quantitative caspase-3 protein changes in ovarian cancer in different periods, the apoptosis in ovarian cancer in different periods was studied. By rays irradiation, and then the number of cells was counted, in order to study different types of ovarian cancer cells radiation sensitivity. Results: Immunohistochemistry, PCR, Western Blot, results with ovarian cells canceration showed that survival bcl-2 genes of apoptosis was activated, lethal gene bax of apoptosis was inhibited, the apoptosis protein caspase-3 declined. At the same time, rays irradiation and statistical results showed that the different types of ovarian cancer cells to radiation sensitivity were different. Conclusion: Caspase-3 protein quantitied with ovarian canceration is declining, but for ovarian cancer cells, the more caspase-3 protein is, the stronger radiation sensitivity is.
