经血管周围移植BMSCs抑制腹主动脉瘤生长

摘要:

目的:通过血管周围途径移植骨髓间充质干细胞(bone mesenchymal stem cells,BMSCs),观察其能否抑制腹主动脉瘤的生长。方法:我们采取全骨髓贴壁的方法分离BMSCs,进行体外扩增、纯化,并采取流式细胞术进行鉴定。利用弹性蛋白酶灌注的方法建立大鼠腹主动脉瘤模型,以Pluronic F-127为载体将BMSCs移植于动脉瘤周围,通过测量血管直径、病理切片染色、PCR等方法评估不同组腹主动脉瘤的生长。结果:经过BMSCs治疗的实验组与对照组相比,动脉瘤形成的平均直径明显缩小,MMP9的表达降低而TIMP的表达升高,且血管平滑肌细胞的数量明显增多。结论:以Pluronic F-127为载体将BMSCs移植于腹主动脉瘤周围,可以成功抑制腹主动脉瘤的生长。

Objective:To explore the possibility of controlling elastase-induced abdominal aortic aneurysm progression by bone mesenchymal stem cells(BMSCs) transplanted to the periadventitial side of the aneurysm wall. Methods:Rat bone marrow mesenchymal stem cells were isolated from the bilateral tibial and femoral bones by the method of whole bone marrow adherence, then they were purified and passaged in vitro. Abdominal aortic aneurysm models were builted by the method of elastase perfusion.BMSCs were transplanted to the periadventitial side of the aneurysm wall by the carrier of Pluronic F-127, aneurysm progression was assessed by measuring the diameter of abdominal aorta, tissue staining, real-time PCR. Results:By contrast of control group, mean aneurysm diameter was narrowed, gene expression level of MMP9 was reduced, expression level of TIMP was increased, and the vascular smooth muscle cells were notably increased in experimental group. Conclusion:Abdominal aortic aneurysm progression can be successfully attenuated by bone mesenchymal stem cells transplanted to the periadventitial side of aneurysml by the carrier of Pluronic F-127.
