载阿霉素的磁性微泡双模式的成像效果及其对淋巴瘤细胞毒性的研究

摘要:

目的:探讨多功能高分子微泡(MPMBs)双模式成像效能及其联合超声对DLBCL弥漫大B细胞淋巴瘤(DLBCL)细胞株SU-DHL-4增殖和凋亡的影响.方法:以机械振荡法制备共载阿霉素及超顺磁性氧化铁的MPMBs,观察其体外增强B超、磁共振显影的能力;将SU-DHL-4细胞株按处理方法不同分组,CCK-8法检测各组细胞活力,吖啶橙/溴化乙啶(AO/EB)双染法观察细胞形态.结果:MPMBs较普通高分子微泡(PMBs)具有更好的超声、磁共振显影效果;MPMBs联合超声对DLBCL细胞增殖的抑制强于其他处理组.结论:MPMBs是一种可集治疗与超声、磁共振成像于一体的多功能造影剂.

Objective: To investigate the dual-mode imaging ability of multifunctional polymer microbubbles(MPMBs), and to observe its effect on cell proliferation and apoptosis in SU-DHL-4 cells.Methods: The MPMBs were made by mechanical vibration method. Enhancement of ultrasound (US)/magnetic resonance(MR) imaging were evaluated in vitro. The SU-DHL-4 cells were divided into several groups according to different treatments, then the viability of cells were measured by CCK-8 assay and the morphologic changes were detected by AO/EB staining.Results: US/MR dual-mode imaging showed its better property compared with ordinary polymer microbubbles(PMBs). Besides, anti-proliferation effect of MPMBs combined US on cells was much better than other treatment groups. Conclusion: The MPMBs can be served as a mutifunctional imaging agent for treatment and imaging.
