低氧预处理对大鼠心肌成纤维细胞NLRP3基因表达的影响

摘要:

目的:研究NLRP3基因在缺氧大鼠心肌成纤维细胞中的表达。方法:将大鼠心肌成纤维细胞体外原代培养,以氯化钴和低氧模拟心肌缺氧刺激,利用Real-time PCR和Western blot检测NLRP3基因在大鼠心肌成纤维细胞中的表达。结果:成功地体外培养出鼠心肌成纤维细胞,NLRP3基因在大鼠心肌成纤维细胞缺氧状态下表达明显增高。结论:NLRP3基因在大鼠心肌成纤维细胞缺氧状态下表达上调可以作为心肌梗死发生评判的重要因子。

Objective: To investigate expression of NLRP3 gene after hypoxic preconditioning in rat cardiac fibroblasts. Methods: Rat cardiac fibroblast cells were cultured in vitro. After treated with certain concentration of CoCl2 or hypoxia-mimicked myocardial infarction, expression of NLRP3 gene was detected by Real-time PCR and Western blot in rat cardiac fibroblasts. Results: Rat cardiac fibroblasts cells model had been successfully established. The expression of NLRP3 gene significantly increased under CoCl2-mimicked hypoxia in rat cardiac fibroblasts. Conclusion: NLRP3 gene is up-regulated after hypoxic preconditioning in rat cardiac fibroblasts, and may be a significant contributor to myocardial infarction development.
