Objective To observe dynamical changes of injured airway by way of establishing rat models of chronic obstructive pulmonary disease (COPD).Methods The rats were intratracheally instilled with lipopolysaccha- ride(LPS) twice and exposed to cigarette smoke to make COPD models,whose morphological changes of airways and lung tissue were dynamically observed; interleukin-8(IL-8) in the serum and bronchoalveolar lavage fluid (BALF) were detected. Results After the model are established,the epithelial cilia cohered,partially fell off;the amount of macrophage lysosome in pulmonary alveolus lumen increased and emphysema was noted;mean lung internal lining interval(MLI)significantly increased, mean alveolar number(MAN) decreased significantly, IL-8 in serum and BALF increased significantly.Conclusion COPD rat models are successfully established by passive cigarette smoking plus intratracheal instillation of LPS,and their degree of inflammation in the airway is correlated with its morphological change. |
