Objective To investigate the possibility for acellular dermis(AD) prepared by trypsase and repeated freeze thaw cycles to be an ideal dermal substitute for construction of composite skin.Methods The acellullar xeno dermal matrixes(2?cm×2?cm)were implanted under epidermis of SD rat.2, 4, 6 and 8 weeks after implantation,the size of the implanted AD was measured and an excisional biopsy of each implant was obtained for histological examination.Results Four weeks after implanting,the structure of basement memberane presented normally,and the structure of collagen changed little.Some fibroblasts and microvessel and no inflammatory cells develop evidently between collagens.Conclusion AD prepared by trypsase and repeated freeze thaw cycles is a kind of ideal dermal substitute,which can induce ingrowth of microvessels and fibroblasts,and provides nutrition for the above cultured epidermis and the overlap of autograft. |
