高效液相色谱法测定当归中阿魏酸含量方法的探讨

摘要：

目的:探讨阿魏酸稳定性及高效液相色谱法测定当归中阿魏酸含量的方法.方法:以甲酸∶水(5∶95)为阿魏酸对照品的溶剂,甲酸∶甲醇(5∶95)为样品的溶剂,固定相为shim-pack CLC-ODS,流动相为0.2%甲醇乙腈∶0.1%磷酸0.1%三乙胺水溶液(20∶80),检测波长为324 nm,流速为1 ml*min-1.结果:阿魏酸在0.704～17.6 mg*L-1范围内线性良好(r=0.999 8,n=5),平均加样回收率为99.33%,RSD为0.60%(n＝6).结论:高效液相色谱法简便快速,可用于当归药材的含量测定.

Objectives To improve the stability of ferulic acid and to determine ferulic acid content in angelia sinensis (Oliv.) diels.Methods HCOOH∶H 2O(5∶95) was used as solvant for ferulic acid and HCOOH∶MeOH(5∶95) for the sample.Ferulic acid content was measured by high-performance liquid chromatographic method on shim-pack ODS,using 0.2% 东南大学学报(医学版) J Southeast Univ (Med Sci Edi) 2003,Mar;22(2):101?104MeOH in CH 3CN∶0.1%(C 2H 5) 3N 0.1%H 3PO 4 in H 2O (20∶80) as mobile phase with detecting wavelength of 324?nm.The flow rate was 1?ml·min -1.Results The standard curve of ferulic acid was linear over the range of 0.704～17.6?mg·L -1 with the correlation coefficient of 0.999?8(n=5).The average recovery of ferulic acid was 99.33%.The average variation was less than 0.60%(n=6).Conclusion The method is simple and rapid,which is suitable for the determination of ferulic acid in angelia sinensis (Oliv.) diels.
