Objective To investigate the effect of low dose of potassium iodide(KI) on fibrocystic breast disease (FBD) in modeling SD rats.Method The fibrocystic breast disease model was established by subcutaneous injection of estradiol benzoas 0.5?mg·(kg·d) -1 for 25 days,followed by progesterone 4?mg·(kg·d) -1 for 5 days in female SD rats.After the model was established, the rats were divided into three groups:the model control group (treated by NS),the KI therapy groups (treated by KI 3,9 or 27?mg·(kg·d) -1 and tamoxifen group (TAM)(treated by TAM 12?mg·(kg·d) -1 ).The preventive therapy with KI 9?mg·(kg·d) -1 was performed since the beginning of modeling.The therapeutic effect of KI was evaluated by pathological examination,and the serum levels of estradiol(E 2),estriol(E 3),progesterone and luteinizing hormone(LH) were determined by radioimmunoassay method.Results After administering KI 3,9 or 27?mg·(kg·d) -1 30?d,the pathological changes of the modeling rats were significantly abated.The serum levels of E 2 and E 3 were decreased in a dose dependent way,whereas,the serum levels of progesterone and LH were significantly higher than those of model control rats.The production and development of fibrocystic breast were attenuated by preventive administration of KI in rats.Conclusion KI might be used for therapy and prevention of fibrocystic breast disease. |
