新型融合毒素hIL-15-TNFα表达质粒的构建

摘要:

目的:构建hIL-15-TNFα融合毒素的表达质粒,为该融合毒素的功能研究及临床应用前景的分析评价奠定基础.方法:将hIL-15成熟肽基因与人工改造的TNFα基因按正确的阅读框架融合,定向克隆于表达载体pET16b.结果:经酶切分析及DNA序列分析,所构建的质粒均插入hIL-15-TNFα融合基因.结论:利用pET16b表达载体可成功构建融合毒素hIL-15-TNFα的表达质粒.

Objective  To characterize the biological function and clinical perspective of hIL?15?TNF    α   fusion toxin,a plasmid expressing this fusion molecule was constructed.Method  Artifically modified TNF    α  cDNA was fused precisly to the reading frame of hIL?15 and further cloned into the   E.coli   expression vector pET16b.Results  The recombinant plasmid containing hIL?15?TNF    α   fusion gene was obtained as demonstrated by restriction analysis and DNA sequencing.Conclusion  The recombinant plasmid expressing hIL?15? TNF    α   fusion toxin is successfully constructed.
