人胰腺癌端粒酶亚单位的表达及其意义

摘要:

目的:探讨端粒酶亚单位表达水平与人胰腺癌发生的关系,以及其表达与端粒酶活性高低的相关性.方法:应用RT-PCR、DNA测序等技术研究端粒酶亚单位在胰腺癌中的表达情况,用ELISA法检测端粒酶活性.结果:24例胰腺癌标本中hTERTmRNA表达阳性率为83.3%,而癌旁组织为8.3%,两者有显著性差异.hTERTmRNA的表达是人端粒酶活性表达的关键性因素.hTR及TP1mRNA在癌组织及癌旁组织中的表达无统计学差异,与端粒酶活性的表达无相关性.结论:端粒酶参与了胰腺癌的发生;hTERTmRNA表达水平的上调可能在胰腺癌形成过程中起重要作用.

Objective  To directly investigate the relationship between the telomerase subunit expression and pancreatic carcinogenesis,and whether telomerase subunit is an important component of telomerase or not.Methods  The expression of telomerase subunit was examined by RTpolymerase chain reaction(RTPCR) and DNA sequencing methods.Telomerase activity was detected by ELISA.Results  The analysis of pancreatic carcinoma revealed that telomerase activities were positive in 21 of 24 cases (87.5%),hTERT mRNA in 20 of 24 cases (83.3%),normal tissues were 12.5% and 8.3%.There was a significant difference between the two groups.Our findings showed that the expression of hTR and TP1 mRNA was not correlated with telomerase activity,while the expression of hTERT mRNA was.Conclusions  Our finding shows that the expression of telomerase activity has an relationship with carcinogenesis.A strong correlation was found between telomerase activity and hTERT mRNA expression and the up-regulation of hTERT mRNA expression may play a critical role in human carcinogenesis.
