摘要:

目的:研究三磷酸肌醇受体(IP3R)在大鼠血管平滑肌细胞(VSMC)的表达 .方法:用异硫氰酸胍酸酚一步法提取大鼠VSMC总核糖核酸(RNA ).用AMV逆转录酶反转录,以IP3R各亚型的引物进行反转录PCR(RT PCR),并对扩增产物进行克隆及测序.用间接免疫荧光标记技术检测I型IP3R在VSMC的分布. 结果:3个亚型IP3R mRNA在VSMC中均有表达,扩增片断分别为405、183和169 bp .VSMC中表达缺失型I型IP3R.免疫荧光染色显示,Ⅰ型IP3R在细胞质内围绕核膜周边分布.结论:大鼠血管平滑肌细胞内表达3种亚型的IP3R, 提示单一细胞内存在复杂多样的信号转导途径.

Objective  To investigate the expression of inositol 1,4,5?triphosphate receptors (IP  3R) in cultured vascular smooth muscle cells(VSMC) of rat.Methods  Expression of IP  3R mRNA in VSMC was measured by reverse transcription  polymerase chain reaction (RT  PCR).The amplified PCR products were subcloned and sequenced.The immunocytochemical analytic technique was used to observe IP  3R expression intracellularly.Results  Three subtypes of IP3R expressed within rat VSMC were identified and the short non  neuronal form of type I IP  3R was detected.Immunofluorescence staining indicated that the IP  3R was preferentially located at the peri  nuclear membrane in cellular plasma.Conclusion  The present study confirms that three subgroup of IP  3R are expressed in VSMC,and it suggests that IP  3 signal transduction pathway within a single cell be of heterogeneity.
