不同剂量抗纤复方药物血清对正常大鼠肝细胞增殖与活力的影响

摘要：

目的：探讨大鼠给予不同剂量抗纤复方药物后其血清对体外培养正常大鼠肝细胞增殖与活力的影响。方法：常规培养大鼠肝细胞，用5、10与20倍剂量抗纤复方药物血清分别温育肝细胞48h，同时以对照血清作为参照，用[3H]-TdR掺入法测定细胞增殖，[3H]-Pro掺入法测定细胞活力。结果：各剂量组抗纤复方药物血清均能增加肝细胞[3H]-TdR(20%的20倍剂量组除外)和[3H]-Pro掺入量，其中10%的10倍剂量抗纤复方药物血清作用最满意。结论：抗纤复方药物血清能促进正常肝细胞增殖和活力，最佳剂量为10倍剂量。

Objective To study the effect of drug sera of anti fibrosis compond priscription (ACP) on the functions of primarily cultured normal rat hepatocyte such as proliferation and viability.Methods Primarily cultured normal rat hepatocytes were cultured with regular method, and incubated with “five twenty times” drug sera of ACP for 48?h.The cell proliferation was measured by [ 3H] TdR incorporation, cell viability by [ 3H] Pro incorporation.Results The three drug sera groups could promote the proliferation and viability of normal rat hepatocytes,10%“ten fold” drug sera of ACP showed the best effect.Conclusion The drug sera of ACP could enhance the proliferation and viability of normal rat hepatocytes,the best drug concentration is “ten fold” drug sera of ACP in vitro .
