大鼠成骨细胞的体外培养和生物学特性的研究

摘要:

目的:建立一种成骨细胞的体外培养方法,并研究不同培养时期成骨细胞的生物学特性.方法:选用成年雌性大鼠松质骨为培养细胞来源,组织块培养法和胰酶消化法相结合进行成骨细胞培养,并测定成骨细胞不同培养阶段的碱性磷酸酶(AKP)活性和骨钙素(OC)分泌水平.结果:体外培养所获成骨细胞纯度高、数量多,且培养时间明显缩短.培养细胞近融合时,产生的AKP和OC处于低水平,融合后3 d、14 d AKP和OC的产生明显增加.结论:实验采用的改良大鼠成骨细胞培养方法较为方便、切实可行.体外培养的大鼠成骨细胞在不同生长阶段,其骨基质蛋白的产生发生相应变化.

Objective  This study aimed at establishing a modified culture model of normal rat osteoblast and researching on the biologic characteristic of osteoblast during different culture periods.Method  When bone tissue culture combined with trypsin digestion was used,lots osteoblast?like cells from adult female rat trabecular bone were obtained.Cellular alkline phosphatase(AKP) and osteocalcin(OC) in the culture medium were assayed.Result  A great number of osteoblastic?like cells had been cultured with higher purity and less time in our experiment.The secretion of AKP and OC was found to be in a low level shortly before the confluence of the cultured cells.The production of AKP and OC was increased at day  3 and 14 after confluence.Conclusion  The culture of rat osteoblast   in vitro   has different growth stages,accompanying with a changing production of bone matrix protein.
