凋亡相关蛋白BCL-2与白血病化疗药物敏感性的研究

摘要:

目的:研究急性白血病(AL)细胞BCL-2表达及其与体外化疗药物敏感性的关系.方法:采用Western blotting技术和四甲基偶氮唑盐比色法(MTT法),分别检测HL-60、U937细胞株和6例急性髓性细胞白血病(AML)细胞BCL-2表达水平及体外对柔红霉素(DNR)的敏感性.结果:(1)BCL-2表达水平HL-60细胞高于U937细胞,而6例AML细胞BCL-2表达水平不同;(2)MTT检测结果显示,DNR对HL-60抑制率低于U937;BCL-2高表达的AML细胞,DNR抑制率低.结论:细胞系和白血病原代细胞的BCL-2高表达伴低药敏与AML疗效有关.

Objective  To investigate the relationship between the expression of BCL?2 protein and the  chemosensitivity of acute leukemic cells   in vitro  .Method  The levels of BCL?2 protein and susceptibility to DNR(daunorubicin) in HL?60 cells,U    937   cell lines and the bone  marrow cells of six cases of AML were detected by the method of Western blotting and MTT assay.Results  (1) The HL?60 cells have a higher BCL?2 protein expression than U    937   cells.All of the six cases of AML have different levels of BCL?2 protein;(2) The U    937   cells are more susceptible to DNR than HL?60,while in AML cases,the higher levels of BCL?2 protein were associated with poor susceptibility. Conclusion  High levels of BCL?2 protein may associate with poor response to chemical therapy.
