人瘢痕成纤维细胞中Sp-1等转录因子的研究

摘要:

目的:研究增生性瘢痕成纤维细胞中纤维化相关的转录因子.方法:人皮肤增生性瘢痕成纤维细胞原代、传代培养后抽提核蛋白.人工法合成转录因子Sp-1、NF-Ⅰ、NF-кB识别序列的双链寡核苷酸,并采用Dig-ddUTP3'末端标记该双链寡核苷酸.电泳迁移率改变实验(EMSA)法研究瘢痕成纤维细胞中纤维化相关的Sp-1、NF-Ⅰ、NF-кB等转录因子.结果:在人增生性瘢痕成纤维细胞的核蛋白中存有纤维化相关的Sp-1、NF-Ⅰ、NF-κB等转录因子.结论:瘢痕纤维化的发生、发展与纤维化相关的转录因子SSp-1、NF-Ⅰ、NF-кB密切相关.

Objective  To analyze the transcription factors r elated to fibrosis  in human hypertrophic scar fibroblasts. Methods  Fibroblasts ob tained from the  hypertrophic scar of a 3-year-old patient were primarily cultured and subcultu red  in vitro, and then nucleoproteins were extracted from the cells. The  double-st randed oligonucleotides  specifically  recognized by transcription factors Sp-1 ,  NF-Ⅰ, NF-κB were synthesized and end-labeled with Dig-ddUTP, then used a s a pr obe in electrophoretic mobility shift assays(EMSA) to identify the transcription  factors related to fibrosis in hypertrophic scar fibroblasts.  Result   The data  confirmed that there were  transcription factors Sp-1, NF-Ⅰ, NF-κB in the  hype rtrophic scar fibroblasts. Conclusion  Hypertrophic scar develop ing  is related to transcription factors Sp-1, NF-Ⅰ, NF-κB.
