Objective To analyze the transcription factors r elated to fibrosis in human hypertrophic scar fibroblasts. Methods Fibroblasts ob tained from the hypertrophic scar of a 3-year-old patient were primarily cultured and subcultu red in vitro, and then nucleoproteins were extracted from the cells. The double-st randed oligonucleotides specifically recognized by transcription factors Sp-1 , NF-Ⅰ, NF-κB were synthesized and end-labeled with Dig-ddUTP, then used a s a pr obe in electrophoretic mobility shift assays(EMSA) to identify the transcription factors related to fibrosis in hypertrophic scar fibroblasts. Result The data confirmed that there were transcription factors Sp-1, NF-Ⅰ, NF-κB in the hype rtrophic scar fibroblasts. Conclusion Hypertrophic scar develop ing is related to transcription factors Sp-1, NF-Ⅰ, NF-κB. |
