大鼠多发性脑梗死后脑组织内皮细胞ICAM-1表达的变化

摘要:

目的:观察大鼠多发性脑梗死后脑组织内皮细胞表达细胞间粘附分子-1(ICAM-1)蛋白的时程变化.方法:48只健康雄性SD大鼠制成多发性脑梗死模型,梗死后1 h~14 d处死并取脑,用免疫组织化学法标记脑组织切片ICAM-1蛋白,另外6只雄性大鼠作为对照.结果:多发性脑梗死后1 h脑组织内皮细胞即有ICAM-1蛋白表达,2 h达高峰,然后表达下降,持续表达 7 d,14 d时无明显表达.结论:支持内皮细胞ICAM-1表达增加是其对缺血-再灌注的一种自身反应的假设.

Objective   To investigate the changes of ICAM、1 protein expression of ischemia brain induced by embolization in rats. Methods  Male SD rats (n=48) were subjected to multi-focal cerebral infarct, brain tissues were obtained at 1、h to 14、d accordingly after the onset of embolization,6 additional control animals were employed as controls. Immunohistochemical method was used to identify specific cell types expressing ICAM、1 protein in brain sections. Results  ICAM、1 protein was detected at 1、h after the onset, maximized at 2、h, and persisted at a low level to 7、d, fell down to baseline by 14、d. Conclusion  This study supports the hypothesis that the endothelium itself upregulates ICAM、1 in response to the stimulus of ischemia-reperfusion.
