HPLC-FD法检测尿儿茶酚胺

摘要:

目的:建立一种简便、灵敏度高、重复性好的尿儿茶酚胺检测方法.方法:应用高效液相色谱(HPLC)系统和荧光检测器,探索检测24h尿去甲肾上腺素、肾上腺素和多巴胺含量的最佳条件以及回收率、线性范围、精密度、最低检测线.结果:流动相甲醇与0.02mol*L-1磷酸二氢钾的比为2∶98、pH值为3.0时测定效果最佳.去甲肾上腺素、肾上腺素和多巴胺回收率依次为69%~78%、63%~73%及60%~71%,最低检测线依次为0.015、0.008、0.003μmol*L-1,RSD为4.9%~10.6%.结论:HPLC-FD法用于临床检测尿儿茶酚胺简便易行,灵敏度高,重复性好,易于临床推广.

Objective  To establish a new method determining catecholamines in urine. Methods  Conditions of determining norepinephrine (NE),epinephrine(E) and dopamine(DA) in 24-h urine were compared by high performance liquid chromatography-fluorescence detection,and the lowest detectable limit,the precision,the rate recovery were detected. Results  The best result was obtained when fluid phase was methanol0.02-mol·L    -1 KH  2PO  4=298 and pH=3.0.The recovery rates of NE-E-DA were 69%-78%-63%-73%-60%-71%,the lowest detectable limit were 0.015,0.008,0.003-μmol·L    -1.RSD is 4.9%-10.6%.Conclusion  The simple and highly sensitive method may be repeated in the clinical detection of CA levels.
