γ-干扰素调控慢性B淋巴细胞白血病B细胞B7.2分子表达

摘要:

目的:研究观察γ-干扰素调控慢性B淋巴细胞白血病(BCLL)B细胞B7.2分子的表达,并观察B7.2水平的变化对免疫反应的影响.方法:用500IU*ml-1浓度的γ-干扰素体外孵育BCLL患者外周血B细胞48h,流式细胞仪检测孵育前后B7.2表达的百分率,电镜观察其形态变化;采用单向混合淋巴细胞反应(MLR)技术观察B7.2水平的变化对淋巴细胞增殖的影响.结果:BCLL患者外周血B细胞经500IU*ml-1浓度的γ-干扰素体外孵育48h后,B7.2的表达从17%上升至21%;电镜观察,细胞未出现明显的树突状细胞的特征性改变;MLR观察,B7.2上调之后,实验组每分钟计数值增高,比较差异有显著性(P<0.05).结论:γ-干扰素可轻度上调BCLL B细胞B7.2分子的表达水平,从而可能提高机体对BCLL细胞的免疫清除.

Objective  To investigate effects of γ-interferon on B7.2 expression in B cells of B cell chronic lymphocytic leukemia and the impact of latter on immune-triggering.Methods  Peripheral blood mononuclear cells (PBSC) were isolated from 8 patients with BCLL (stage Ⅳ) and cultured for 6 hours in vitro,followed by incubation with 500-IU·ml    -1 γ-interferon (γ-IFN) for 48 hours,cells of BCLL were harvested for FCM scan and electron microscope observing. The influence of different B7.2 expression level on lymphocytes proliferation was tested by mixed lymphocyte reaction (MLR).Results  After being incubated with 500-IU·ml    -1 γ-interferon for 48 hours,B7.2 expressed increasingly from 17% to 21% in an average,however,they did not demonstrate the phenotype of dendritic cell.MLR showed CPM value of γ-interferon group increased markedly with a significant difference compared with control group.Conclusion  γ-interferon induces B7.2 expression of B cells of BCLL,whose up-regulation may immunological promote the clearance of leukemia cells.
