Objective To investigate the efficacy of romurtide(RM),the effective component of Bacillus Calmette?Guerin (BCG) in cytokine production by human peripheral blood mononuclear cells (HPBMCs).Methods Different concentrations of romurtide were administered to HPBMCs cultured in vitro for 72 hours, from which the cultured supernate was harvested, and the level of IL?12,IL?4 and GM?CSF were detected by ELISA.Results RM could significantly stimulate HPBMCs to produce both IL?12 and GM?CSF,Th1?associated cytokines in a dose-dependent manner. However,RM had no obvious effect on the secretion of IL?4,the Th2?associated cytokine. Conclusion RM might be an effective adjuvant to induce cellular immune response via the secreation of IL?12 and GM?CSF of human peripheral blood mononuclear cells, suggesting that romurtide might be used for bladder cancer immunotherapy. |
