下丘脑室旁核微量注射P物质对豚鼠呼吸的影响

摘要:

目的:探讨下丘脑室旁核(paraventricular nuclei,PVN)微量注射P物质(substance P,SP)对豚鼠呼吸运动的影响及其作用机制.方法:采用中枢立体定位技术,在豚鼠PVN预先植入带芯的不锈钢外套管.实验时切断双侧迷走神经,自主呼吸,通过内管恒速注射不同药物,持续记录呼吸流量、动脉血压、膈肌肌电活动.结果:与生理盐水组比较,微量注射SP后2~27min,膈肌肌电活动增强,通气量增加(均为P<0.001).微量注射SP后3~20min,血压升高(P<0.05).若先以SP的受体拮抗剂[D-Pro2,D-Trp7,9]-SP(DPDTDT)或血管升压素(vasopressin,VP)的V1受体拮抗剂[deamino-Pen1,Val4,D-arg8]-VP(DPVAVP)预处理,则SP的呼吸和血压效应消失.单独注射DPDTDT或DPVAVP对呼吸和血压没有显著影响.结论:PVN内微量注射SP有呼吸兴奋作用,此作用可能是由NK1受体或V1受体介导的.

Objective  TO investigate the effects and mechanisms of microinjection of substance P(SP) into the hypothalamic paraventricular nuclei (PVN) on respiration in guinea pigs.Methods   Experiments were carried out on 36 anesthetized guinea pigs of both sexes,vagotomized and spontaneously breathing,with stainless steels implanted into PVN 24 h before.The drugs were injected into left PVN via the steels,then the effects of respiration and arterial blood pressure were observed for 40?min.Results  Compared with NS,SP enhanced both respiration during 2~27?min (  P  <0.001) and blood pressure during 3~20?min (  P  <0.05).Pretreatment with NK  1 antagonist (DPDTDT) of SP or V  1 antagonist (DPVAVP) of vasopression,SP had no such effects.Neither DPDTDT nor DPVAVP singly had such effects.Conclusion  Microinjection of SP into PVN induced respiratory excitement which was likely obtained via NK  1 and (or) V  1 receptors.
