家兔第四脑室注射P物质对呼吸活动的影响及其与组胺能系统的关系

摘要:

目的:研究脑内P物质(subtance P,SP)对呼吸活动的影响及其与组胺能系统的关系.方法:采用第四脑室微量注射方法,观察家兔第四脑室注射组胺受体阻断剂后SP对呼吸的效应.结果:将SP1.67μg(50μl)注入第四脑室引起呼吸幅度加深,与生理盐水(NS)对照组相比,差异有统计学意义(P<0.01).呼吸频率无明显变化.若用SP受体阻断剂[D-Pro2,D-Trp7,9]-substance P(DPDTDT)8μg预处理,则第四脑室注入SP所致的呼吸兴奋效应消失.若用组胺H1受体阻断剂扑尔敏预处理,则SP引起的呼吸幅度变化与单独给SP组相比,差异有统计学意义(P<0.01).若预先向第四脑室注入组胺H2受体阻断剂西米替丁,则SP引起的呼吸幅度变化与单独给SP组相比,差异无统计学意义(P>0.05).即DPDTDT及扑尔敏均能阻断SP的呼吸增强效应.结论:第四脑室注入SP可引起呼吸增强效应.中枢组胺能通路通过组胺H1受体参与SP的呼吸增强效应.

Objective   To investigate the effect of substance P(SP) on respiration and the relationship between SP and histaminergic system.Methods The method of microinjection in the fourth ventricle was applied to investigate the effect of SP on respiration after pretreating with histamin receptor antagonist.Results  Injection of SP (  1.67?μg  ,50?μl) into the fourth ventricle significantly increased the range of respiration (  P  <0.01) compared to the saline controls.The variation of respiratory frequency was not significant.Pretreating with SP receptor antagonist [D  pro  2,D?Trp    7,9  ]?SP(DPDTDT) 8?μg,the exciting effect of SP on respiration disappeared.Pretreating with H  1 receptor antagonist chlorpheniramine,the excitement of SP on respiration was significant compared to SP controls(  P  <0.01).Injecting SP after pretreatment of H  2 receptor antagonist cimetidine,SP excited respiration insignificantly compared to SP controls(  P  >0.05).Conclusion  Injection of SP (1.67?μg,50?μl) into the fourth ventricle increases the respiratory range mediated by central histaminergic system via H  1 receptor.
