Prx 1高表达对人PC3前列腺癌细胞氧化损伤的作用

摘要:

目的:探讨过氧化物还原酶1(Prx1)表达增加对人肿瘤细胞抗过氧化氢毒性作用的影响.方法:构建Prx1真核表达质粒,稳定转染培养的人PC3前列腺癌细胞,用Western blot检测Prx1在稳定转染PC3细胞中的表达,用MTY法观察细胞的存活率.结果:Western blot分析表明,Prx1真核表达质粒稳定转染的PC3细胞表达Prx 1显著增加;MTT分析表明,在过氧化氢浓度为0.25、0.5、1和2 mmol·L-1时,Prx1稳定转染的PC3细胞存活率显著增加.结论:Prx1高表达显著增强人PC3前列腺癌细胞抗过氧化氢毒性作用.

Objective To investigate the effect of peroxiredoxin 1(Prx?1) overexpression on resistance of human tumor cells to hydrogen peroxide.Methods pcDNA/Prx?1 plasmid was constructed and stably transfected into human PC3 prostate cancer cells.Western blot analysis was used to monitor expression of peroxiredoxin 1 in the stable transfectants.MTT assay was used to observe survival rate of the stable transfectants.Results Compared with pcDNA3 vector,there was significant increase of peroxiredoxin 1 expression in the PC3 cells stably transfected with pcDNA3/Prx 1 monitored by Western blot analysis.When treated with 0.25,0.5,1 and 2?mmol·L^{-1} of hydrogen peroxide,the PC3 cells stably transfected with pcDNA3/Prx 1 increased suvival span.Conclusion Overexpression of Prx 1 obviously increases resistance of human PC3 prostate cancer cells to hydrogen peroxide.
