Objective To observe the effect of ethacridine on growth and expression of MMP?2 and MMP?9 of B16 melanoma cells.Methods After B16 cells were treated with various concentrations of ethacridine,inhibiting ratio of the growth was measured by MTT assay.The expression of MMP-2 protein was detected by ELISA.And then the expression of MMP-2,MMP-9 proteins was detected by immunocytochemistry.Results Proliferation of B16 melanoma cells in vitro was significantly inhibited(P<0.01),showing a time-dependent and dose-dependent manner.MMP-2 secretion was significantly decreased in supernatant secreted by melanoma cells treated with?higher concentrations of ethacridine(6.25,12.5 μg·ml^{-1},P<0.05).The increase of the expression of MMP-2 and MMP-9 proteins attenuated along with the increase of the concentration of ethacridine.Conclusion Ethacridine can markedly inhibit proliferation of B16 melanoma cells cultured in vitro and restrain their metastasis to some extent. |
